Isolation of Subcellular Granules from Second-Stage Juveniles of Meloidogyne incognita.

چکیده
Subcellular granules from the second-stage (preparasitic) juveniles of root-knot nematode Meloidogyne incognita were isolated by isopycnic centrifugation on Percoll. The granules had an apparent density of 1.13 g/cm(3). The relative specific activity of acid phosphatase in the granule extract was 8.4. Acid phosphatase activity was also detected histochemically in the subventral gland granules. Electron microscopy and malate dehydrogenase activity indicated that contamination of granules by mitochondria was negligible. Electrophoresis of the granule extract in the presence of sodium dodecyl sulfate showed 15-20 major protein bands.

مقایسه مولکولی جمعیتهایی از Meloidogyne javanica و Meloidogyne incognita در ایران با روش PCR – RFLP
To carry out this study, total DNA was extracted from eggs and from second stage juveniles of several populations of Meloidogyne javanica and Meloidogyne incognita, using phenol / chloroform method. Following extraction, DNA was electrophoresed on 1% agarose gel to determine its quality and quantity. A specific primer pair (C2F3 / 1108 23 and 20 nucleotides, respectively) was used to discrimina...

متن کاملExtremely Sensitive Thermotaxis of the Nematode Meloidogyne incognita.
Eggs of the root-knot nematode Meloidogyne incognita were acclimated to 23 C. Newly hatched second-stage juveniles migrated toward higher temperatures when placed in shallow thermal gradients averaging 23 C. The threshold gradient for this response was below 0.001 C/cm, with a best estimate of 4 x 10 C/cm. Calculations of physical limitations on thermotaxis indicate that this sensitivity is wel...
متن کاملHost–parasite relationships of Meloidogyne incognita on spinach
Host–parasite relationships in root-knot disease of spinach caused by Meloidogyne incognita race 1 were studied under glasshouse conditions. Nematode-induced mature galls were large and usually contained one or more females and egg masses with eggs. Feeding sites were characterized by the development of giant cells containing granular cytoplasm and many hypertrophied nuclei. The cytoplasm in th...
متن کاملStage-specific Differences in Lectin Binding to the Surface of Anguina tritici and Meloidogyne incognita.
The occurrence and distribution of several lectin binding sites on the outer surfaces of eggs, preparasitic second-stage juveniles (J2), parasitic second-stage juveniles (PJ2), females, and males of two tylenchid nematodes, Anguina tritici and Meloidogyne incognita race 3, were compared. In both species, a greater variety of lectins bound to the eggs than to other life stages; lectin binding to...
